When dissolving (reconstituting) a freeze-dried peptide, the choice of liquid is not a side issue: it partly determines how stable the solution remains and how reliable an experiment is. In practice, three options come up most often: bacteriostatic water, sterile water and — for hard-to-dissolve peptides — a mildly acidic solution such as acetic acid. This guide explains in plain language what the difference is and when researchers choose which.

Bacteriostatic water — the standard choice

Bacteriostatic water is simply water with 0.9% benzyl alcohol added. That benzyl alcohol inhibits the growth of bacteria, so a reconstituted vial can be sampled over several days without micro-organisms quickly growing in it. For most research peptides it is therefore the standard reconstitution liquid: good dissolving capacity and a longer usable life of the solution. You will find it as bacteriostatic water with the accessories.

Sterile water — for short-term use

Sterile water is pure water without a preservative. It is usable when a solution is used immediately or when benzyl alcohol is undesirable for experimental reasons (for example because it can interfere with an assay). The downside is the shorter usable life: without a preservative the solution is more susceptible to microbial growth and is therefore used or frozen sooner. See sterile water.

Acetic acid — for hard-to-dissolve peptides

Some peptides dissolve poorly in neutral water, often because they tend to clump together (aggregate). For those cases the literature mentions a mildly acidic solution such as dilute acetic acid: the lower pH can help get the peptide into solution. How do you choose? — a simple rule of thumb

  • Standard, multi-day use: bacteriostatic water.
  • Immediate use or benzyl-alcohol-free needed: sterile water.
  • Peptide dissolves poorly / clumps: a mildly acidic solution (e.g. dilute acetic acid), depending on the protocol.
